how to tune a ukulele with a chromatic tuner?

Hi, I have a ukulele concert just bought about a week ago. I am new to the whole thing. I was wondering if LAEC basic C tuning is tuned to 440Hz, so I need to change it if I wanted to set it to B tuning? or can I keep the same and only the melody is a half step down? I also have another question. I just tried to adjust my ukulele (which I have named Pablo) with the AP program online chromatic guitar tuner GCE strings tuned well, but the string will not tune right. The sequence is very flexible, but the tuner says it is the right note. It is the sequence a supposed to be released like that? let me know! Thanks guys
